Leica TCS SP5

The Leica TCS SP5 confocal set-up is built around an inverted DMI 6000 B microscope with motorized stage. The Tandem scan-head can be used at a defined frequency or at a much higher resonant frequency. The first mode yields high quality images in a wide field of view and allows a static „beam park“ function (e.g. for FRAP analysis). With the resonant scanner time-resolved measurements can be done in a true confocal point-scanning way, recording up to 25 images/sec of 512x512 pixels. In combination with the piezo-driven objective stage fast recording of z-stacks is possible for 3D particle tracking applications, typically using 0.25s for a 20 image stack of 128x128 pixels. The scanner is equipped with three detectors for simultaneous detection of different fluorescent dyes and, in addition, images of the transmitted light can be taken. Furthermore, the LAS MicroLab software offers FRET and FRAP wizards.
